Cryopreservation of Protoplast-Derived Cell Suspensions of Rice (Oryza sativa L.)

Abstract: The protoplast-derived cell suspensions of rice (Oryza sativa L. cv. A 58 MS) were frozen in liquid nitrogen (LN) in the presence of 10–20% dimethylsulfoxide (DMSO) and 10–20% sucrose in combination, the survival reached 40-50% of the control. The retrieved cells were proliferated in Linsmier-Skoog (LS, 1965) medium supplemented with 2×10-5 mol/ l 2,4-D. Shoots and roots were induced from calli formed from frozen cells in LS medium with l×l0 mol/l NAA plus 4×10-6 mol/l kinetin and lxl0-6mol/l 2 IP. The results are discussed.
