Microdissection and PCR Amplification of Rye Chromosome

Abstract: The 18 ( 14A + 4B) chromosomes of rye ( Secale cereale L. ) were isolated by microdissection technique and the chromosomal DNA was amplified using the artificial synthetic single unique oligonucleotide-primed polymerase chain reaction (SUP-PCR). Southern hybridization demonstrated that the SUP-PCR products have homology with rye genome DNA.
